Inheritance of duplicated fumarase and phosphoglucomutase loci in lake trout (Salvelinus namaycush)

Abstract

Two polymorphic, duplicated loci (fumarase-1,2 and phosphoglucomutase-3,4), previously undescribed in salmonids, were examined in lake trout to determine their electrophoretic expression, mode of inheritance, and linkage associations. Both loci appear to be located on chromosomes which show residual tetrasomy. Neither locus pair shows joint segregation with other loci examined or with each other.

Breeder turnover creates allelic variation in groups of gray wolves

Genetic diversity is an important driver affecting the health of wildlife populations. In cooperatively breeding species, human impacts and breeder turnover can affect genetic diversity in groups. We generally do not have strong inferences about how the genetic composition of a group changes through time as individuals are lost (e.g., die, emigrate) or adopted (e.g., immigrate). I wanted to know how breeder turnover, group size, and harvest affected the fluctuation of unique alleles in groups of gray wolves ( Canis lupus ) in Idaho, USA, during 2008–2020. Turnover of breeding males was strongly associated with allelic change in groups. Turnover of breeding females also had a strong association with allelic change in groups, but was not the most supported model. Harvest was strongly correlated with breeding female turnover but not breeding male turnover. Outside of breeding female turnover, harvest generally had little effect on allelic change in groups. Groups rarely adopted new individuals unless there was a breeding vacancy. I show that over time groups gain and lose alleles in roughly equal proportions, but there are episodic changes to alleles in groups as a function of breeding male turnover. These findings have implications for how we define and evaluate group persistence and breeder lineages in cooperative breeders. Such definitions have important implications for studying the evolution and maintenance of cooperative breeding. It may be beneficial to define characteristics and vital rates of groups based, at least in part, on their underlying genetics when such information can be obtained.

Landscape genetics of a sub-alpine toad: Climate change predicted to induce upward range shifts via asymmetrical migration corridors

Climate change is expected to have a major hydrological impact on the core breeding habitat and migration corridors of many amphibians in the twenty-first century. The Yosemite toad ( Anaxyrus canorus ) is a species of meadow-specializing amphibian endemic to the high-elevation Sierra Nevada Mountains of California. Despite living entirely on federal lands, it has recently faced severe extirpations, yet our understanding of climatic influences on population connectivity is limited. In this study, we used a previously published double-digest RADseq dataset along with numerous remotely sensed habitat features in a landscape genetics framework to answer two primary questions in Yosemite National Park: (1) Which fine-scale climate, topographic, soil, and vegetation features most facilitate meadow connectivity? (2) How is climate change predicted to influence both the magnitude and net asymmetry of genetic migration? We developed an approach for simultaneously modeling multiple toad migration paths, akin to circuit theory, except raw environmental features can be separately considered. Our workflow identified the most likely migration corridors between meadows and used the unique cubist machine learning approach to fit and forecast environmental models of connectivity. We identified the permuted modeling importance of numerous snowpack-related features, such as runoff and groundwater recharge. Our results highlight the importance of considering phylogeographic structure, and asymmetrical migration in landscape genetics. We predict an upward elevational shift for this already high-elevation species, as measured by the net vector of anticipated genetic movement, and a north-eastward shift in species distribution via the network of genetic migration corridors across the park.

Environmental gradients of selection for an alpine-obligate bird, the white-tailed ptarmigan (Lagopus leucura)

The warming climate will expose alpine species adapted to a highly seasonal, harsh environment to novel environmental conditions. A species can shift their distribution, acclimate, or adapt in response to a new climate. Alpine species have little suitable habitat to shift their distribution, and the limits of acclimation will likely be tested by climate change in the long-term. Adaptive genetic variation may provide the raw material for species to adapt to this changing environment. Here, we use a genomic approach to describe adaptive divergence in an alpine-obligate species, the white-tailed ptarmigan ( Lagopus leucura ), a species distributed from Alaska to New Mexico, across an environmentally variable geographic range. Previous work has identified genetic structure and morphological, behavioral, and physiological differences across the species’ range; however, those studies were unable to determine the degree to which adaptive divergence is correlated with local variation in environmental conditions. We used a genome-wide dataset generated from 95 white-tailed ptarmigan distributed throughout the species’ range and genotype–environment association analyses to identify the genetic signature and environmental drivers of local adaptation. We detected associations between multiple environmental gradients and candidate adaptive loci, suggesting ptarmigan populations may be locally adapted to the plant community composition, elevation, local climate, and to the seasonality of the environment. Overall, our results suggest there may be groups within the species’ range with genetic variation that could be essential for adapting to a changing climate and helpful in guiding conservation action.
