Aminobacter ciceronei sp. nov. and Aminobacter lissarensis sp. nov., isolated from various terrestrial environments

Abstract

The bacterial strains IMB-1 T and CC495 T , which are capable of growth on methyl chloride (CH 3 Cl, chloromethane) and methyl bromide (CH 3 Br, bromomethane), were isolated from agricultural soil in California fumigated with CH 3 Br, and woodland soil in Northern Ireland, respectively. Two pesticide-/herbicide-degrading bacteria, strains ER2 and C147, were isolated from agricultural soil in Canada. Strain ER2 degrades N -methyl carbamate insecticides, and strain C147 degrades triazine herbicides widely used in agriculture. On the basis of their morphological, physiological and genotypic characteristics, these four strains are considered to represent two novel species of the genus Aminobacter , for which the names Aminobacter ciceronei sp. nov. (type strain IMB-1 T =ATCC 202197 T =CIP 108660 T =CCUG 50580 T ; strains ER2 and C147) and Aminobacter lissarensis sp. nov. (type strain CC495 T =NCIMB 13798 T =CIP 108661 T =CCUG 50579 T ) are proposed.

Syntrophotalea acetylenivorans sp. nov., a diazotrophic, acetylenotrophic anaerobe isolated from intertidal sediments

A Gram-stain-negative, strictly anaerobic, non-motile, rod-shaped bacterium, designated SFB93 T , was isolated from the intertidal sediments of South San Francisco Bay, located near Palo Alto, CA, USA. SFB93 T was capable of acetylenotrophic and diazotrophic growth, grew at 22–37 °C, pH 6.3–8.5 and in the presence of 10–45 g l −1 NaCl. Phylogenetic analyses based on 16S rRNA gene sequencing showed that SFB93 T represented a member of the genus Syntrophotalea with highest 16S rRNA gene sequence similarities to Syntrophotalea acetylenica DSM 3246 T (96.6 %), Syntrophotalea carbinolica DSM 2380 T (96.5 %), and Syntrophotalea venetiana DSM 2394 T (96.7 %). Genome sequencing revealed a genome size of 3.22 Mbp and a DNA G+C content of 53.4 %. SFB93 T had low genome-wide average nucleotide identity (81–87.5 %) and <70 % digital DNA–DNA hybridization value with other members of the genus Syntrophotalea . The phylogenetic position of SFB93 T within the family Syntrophotaleaceae and as a novel member of the genus Syntrophotalea was confirmed via phylogenetic reconstruction based on concatenated alignments of 92 bacterial core genes. On the basis of the results of phenotypic, genotypic and phylogenetic analyses, a novel species, Syntrophotalea acetylenivorans sp. nov., is proposed, with SFB93 T (=DSM 106009 T =JCM 33327 T =ATCC TSD-118 T ) as the type strain.

Neisseria arctica sp. nov. isolated from nonviable eggs of greater white-fronted geese (Anser albifrons) in Arctic Alaska

During the summers of 2013 and 2014, isolates of a novel Gram-negative coccus in the Neisseria genus were obtained from the contents of nonviable greater white-fronted goose (Anser albifrons) eggs on the Arctic Coastal Plain of Alaska. We used a polyphasic approach to determine whether these isolates represent a novel species. 16S rRNA gene sequences, 23S rRNA gene sequences, and chaperonin 60 gene sequences suggested that these Alaskan isolates are members of a distinct species that is most closely related to Neisseria canis, N. animaloris, and N. shayeganii. Analysis of the rplF gene additionally showed that our isolates are unique and most closely related to N. weaveri. Average nucleotide identity of the whole genome sequence of our type strain was between 71.5% and 74.6% compared to close relatives, further supporting designation as a novel species. Fatty acid methyl ester analysis showed a predominance of C14:0, C16:0, and C16:1ω7c fatty acids. Finally, biochemical characteristics distinguished our isolates from other Neisseria species. The name Neisseria arctica (type strain KH1503T = ATCC TSD-57T = DSM 103136T) is proposed.

Deferrisoma paleochoriense sp. nov., a thermophilic, iron(III)-reducing bacterium from a shallow-water hydrothermal vent in the Mediterranean Sea

A novel thermophilic, anaerobic, mixotrophic bacterium, designated strain MAG-PB1 T , was isolated from a shallow-water hydrothermal vent system in Palaeochori Bay off the coast of the island of Milos, Greece. The cells were Gram-negative, rugose, short rods, approximately 1.0&thinsp;&mu;m long and 0.5&thinsp;&mu;m wide. Strain MAG-PB1 T grew at 30&ndash;70&thinsp;&deg;C (optimum 60&thinsp;&deg;C), 0&ndash;50 g NaCl l &minus;&thinsp;1 (optimum 15&ndash;20 g l &minus;&thinsp;1 ) and pH&thinsp;5.5&ndash;8.0 (optimum pH&thinsp;6.0). Generation time under optimal conditions was 2.5 h. Optimal growth occurred under chemolithoautotrophic conditions with H 2 as the energy source and CO 2 as the carbon source. Fe(III), Mn(IV), arsenate and selenate were used as electron acceptors. Peptone, tryptone, Casamino acids, sucrose, yeast extract, d -fructose, &alpha;- d -glucose and (&thinsp;&minus;&thinsp;)- d -arabinose also served as electron donors. No growth occurred in the presence of lactate or formate. The G+C content of the genomic DNA was 66.7 mol%. Phylogenetic analysis of the 16S rRNA gene sequence indicated that this organism is closely related to Deferrisoma camini , the first species of a recently described genus in the Deltaproteobacteria . Based on the 16S rRNA gene phylogenetic analysis and on physiological, biochemical and structural characteristics, the strain was found to represent a novel species, for which the name Deferrisoma palaeochoriense sp. nov. is proposed. The type strain is MAG-PB1 T (&thinsp;=&thinsp;JCM 30394 T &thinsp;=&thinsp;DSM 29363 T ).
