Transformation of chiral polychlorinated biphenyls (PCBs) in a stream food web

The enantiomeric composition of chiral PCB congeners was determined in Twelvemile Creek (Clemson, SC) to examine potential mechanisms of biotransformation in a stream food web. We measured enantiomeric fractions (EFs) of six PCB atropisomers (PCBs 84, 91, 95, 136, 149, and 174) in surface sediment, fine benthic organic matter (FBOM), coarse particulate organic matter (CPOM), periphyton, Asian clam, mayflies, yellowfin shiner, and semipermeable membrane devices (SPMDs) using gas chromatography (GC-ECD). Nonracemic EFs of PCBs 91, 95, 136, and 149 were measured in almost all samples. Enantiomeric compositions of PCBs 84 and 174 were infrequently detected with racemic EFs measured in samples except for a nonracemic EF of PCB 84 in clams. Nonracemic EFs of PCBs 91, 136, and 149 in SPMDs may be due to desorption of nonracemic residues from FBOM. EFs for some atropisomers were significantly different among FBOM, CPOM, and periphyton, suggesting that their microbial communities have different biotransformation processes. Nonracemic EFs in clams and fish suggest both in vivo biotransformation and uptake of nonracemic residues from their food sources. Pilot- and bench-scale testing of faecal indicator bacteria survival in marine beach sand near point sources

Aim: Factors affecting faecal indicator bacteria (FIB) and pathogen survival/persistence in sand remain largely unstudied. This work elucidates how biological and physical factors affect die-off in beach sand following sewage spills. Methods and Results: Solar disinfection with mechanical mixing was pilot-tested as a disinfection procedure after a large sewage spill in Los Angeles. Effects of solar exposure, mechanical mixing, predation and/or competition, season, and moisture were tested at bench scale. First-order decay constants for Escherichia coli ranged between -0??23 and -1??02 per day, and for enterococci between -0??5 and -1??0 per day. Desiccation was a dominant factor for E. coli but not enterococci inactivation. Effects of season were investigated through a comparison of experimental results from winter, spring, and fall. Conclusions: Moisture was the dominant factor controlling E. coli inactivation kinetics. Initial microbial community and sand temperature were also important factors. Mechanical mixing, common in beach grooming, did not consistently reduce bacterial levels. Significance and Impact of the Study: Inactivation rates are mainly dependent on moisture and high sand temperature. Chlorination was an effective disinfection treatment in sand microcosms inoculated with raw influent. ?? 2009 The Society for Applied Microbiology.

Faecal indicator bacteria enumeration in beach sand: A comparison study of extraction methods in medium to coarse sands

Aims: The absence of standardized methods for quantifying faecal indicator bacteria (FIB) in sand hinders comparison of results across studies. The purpose of the study was to compare methods for extraction of faecal bacteria from sands and recommend a standardized extraction technique. Methods and Results: Twenty-two methods of extracting enterococci and Escherichia coli from sand were evaluated, including multiple permutations of hand shaking, mechanical shaking, blending, sonication, number of rinses, settling time, eluant-to-sand ratio, eluant composition, prefiltration and type of decantation. Tests were performed on sands from California, Florida and Lake Michigan. Most extraction parameters did not significantly affect bacterial enumeration. anova revealed significant effects of eluant composition and blending; with both sodium metaphosphate buffer and blending producing reduced counts. Conclusions: The simplest extraction method that produced the highest FIB recoveries consisted of 2 min of hand shaking in phosphate-buffered saline or deionized water, a 30-s settling time, one-rinse step and a 10 : 1 eluant volume to sand weight ratio. This result was consistent across the sand compositions tested in this study but could vary for other sand types. Bora event variability and the role of air-sea feedback

A two-way interacting high resolution numerical simulation of the Adriatic Sea using the Navy Coastal Ocean Model (NCOM) and Coupled Ocean/ Atmosphere Mesoscale Prediction System (COAMPS??) was conducted to improve forecast momentum and heat flux fields, and to evaluate surface flux field differences for two consecutive bora events during February 2003. (COAMPS?? is a registered trademark of the Naval Research Laboratory.) The strength, mean positions and extensions of the bora jets, and the atmospheric conditions driving them varied considerably between the two events. Bora 1 had 62% stronger heat flux and 51% larger momentum flux than bora 2. The latter displayed much greater diurnal variability characterized by inertial oscillations and the early morning strengthening of a west Adriatic barrier jet, beneath which a stronger west Adriatic ocean current developed. Elsewhere, surface ocean current differences between the two events were directly related to differences in wind stress curl generated by the position and strength of the individual bora jets. The mean heat flux bias was reduced by 72%, and heat flux RMSE reduced by 30% on average at four instrumented over-water sites in the two-way coupled simulation relative to the uncoupled control. Largest reductions in wind stress were found in the bora jets, while the biggest reductions in heat flux were found along the north and west coasts of the Adriatic. In bora 2, SST gradients impacted the wind stress curl along the north and west coasts, and in bora 1 wind stress curl was sensitive to the Istrian front position and strength. The two-way coupled simulation produced diminished surface current speeds of ???12% over the northern Adriatic during both bora compared with a one-way coupled simulation.
